用途:用于真菌的分离培养,还用于一次性使用卫生用品真菌菌落总数检测(GB15979-2002)。

 

原理:蛋白陈提供碳源和氮源;葡萄糖提供能源;琼脂是培养基的凝固剂;氯霉素可抑制细菌的生长。

 

配方(每升):

蛋白胨 10g

葡萄糖40g

琼脂15g

氯霉素 0.1g

最终pH 6.0±0.2

 

使用方法:

 

1、称取沙氏琼脂培养基65.1g,加入蒸馏水或去离子水1L,搅拌加热煮沸至完全溶解,分装三角瓶,115°℃高压灭菌15min,备用。

2、取样液接种5个平皿,每个平皿中加入1mL,然后加入15mL左右已溶化并保温至45℃左右的培养基,摇匀,凝固后,置25±2°C培养7d。

3、分别于3、5、7天观察结果,计算平板上的菌落数,并求平均值。

 

It is used for the isolation and culture of fungi, and also for the detection of total fungal colony counts in disposable hygiene products (GB15979-2002).

 

Principle: Protein hydrolysate provides carbon and nitrogen sources; glucose provides energy; agar is the gelling agent of the medium; chloramphenicol can inhibit the growth of bacteria.

 

Formula (per liter):

Peptone 10g

Glucose 40g

Agar 15g

Chloramphenicol 0.1g

Final pH 6.0 ± 0.2

 

Method of Use:

1. Weigh 65.1g of Sabouraud's agar medium, add it to 1L of distilled water or deionized water, stir and heat to boiling until completely dissolved. Distribute into conical flasks, sterilize at 115°C under high pressure for 15 minutes, and set aside for later use.

2. Take the sample solution and inoculate 5 Petri dishes, adding 1mL to each dish. Then add approximately 15mL of the previously melted and maintained at around 45°C medium, shake evenly, allow to solidify, and incubate at 25±2°C for 7 days.

3. Observe the results on the 3rd, 5th, and 7th days, count the colonies on each plate, and calculate the average.
