用酸溶液溶解试样，溶液中游离的H+用氢氧化钠溶液滴定，以甲基橙-苯胺蓝作为滴定终点指示液，通过消耗的氢氧化钠的量计算出试样中的氨基含量，从而得到脱乙酰基度。
称取于105°C±2°C烘干至恒重的试样0.2g，精确至0.0001g，置于250mL锥形瓶中，加入30mL HCL标准滴定溶液（0.1mol/L），将锥形瓶置于电磁搅拌器座上。在20°C~25°C下搅拌至试样溶解后，加入2滴～3滴甲基橙—苯胺蓝指示液。
常温运输与保存，有效期12个月
The sample is dissolved in an acidic solution, and the free H+ ions in the solution are titrated with sodium hydroxide solution using methyl orange-aniline blue as the indicator for the endpoint. The amino content of the sample is calculated based on the amount of sodium hydroxide consumed, thereby determining the degree of deacetylation.
