1、Folin-酚试剂甲:( 需客户自备或者选用对应的试剂盒)
将1g Na2CO3溶于50mL 0.2moL/L NaOH中,再把0.5g(硫酸铜)CuSO4·5H2O溶于100mL 1%的酒石酸钾钠(或酒石酸钠)溶液,然后将前者50mL与后者1mL混合,现用现配,此试剂只能使用一天,过期失效。
2、Folin-酚试剂乙:
福林酚常用于Lowry 方法进行总蛋白定量
本试剂的浓度为1moL/L,此为Folin-酚试剂应用液。
Folin -酚法测定原理:
在碱性条件下,蛋白质与铜作用生成蛋白质—铜络合物。此络合物将试剂磷钼酸—磷钨酸(FolIn试剂)还原,混合物深蓝色(磷钼蓝和磷钨蓝混合物),颜色深浅与蛋白质含量成正比。此方法操作简便,灵敏度比双缩脲法高100倍,定量范围为5~100μg蛋白质。
操作方法:
1、标准曲线的制作:
取14支试管分成两组,分别加入0、0.1、0.2、0.4、0.6、0.8、1mL标准蛋白质溶液(250μg/mL),用水补足到1mL,加入5mL试剂甲,混匀,于20-25℃放置10分钟,再加入0.5mL试剂乙,立即摇匀,在20-25℃保温30分钟,然后于500nm处比色。测定光密度值,取两组测定的平均值,以蛋白质浓度为横座标,光密度值为纵座标,绘制标准曲线值为定量的依据。
2、样品测定:
取1mL样品溶液(约合20-250μg/多肽或蛋白质)加入5mL试剂甲混匀,于20-25℃放置10分钟,再加0.5mL试剂乙(Folin-酚),立即摇匀,在20-25℃保温30分钟,然后于500nm处比色,以1mL水代替样品作空白对照。测定后,可以在标准曲线中查出未知样品的浓度。若用0.5cm光程的比色杯进行比色,可按下面方法进行操作:取0.2mL样品溶液(约含5-100μg多肽或蛋白质)加入1mL试剂甲(可选用0.3-0.5cm直径的小试管)混匀,10分钟以后,再加入0.1mL试剂乙 ,立即混匀,30分钟后比色。一般来说,若多肽或蛋白质的浓度在2-25μg,测波长755nm,,而25μg以上则采用500nm比色为宜。
注意事项:
1、FolIn试剂显色反应由酪氨酸、色氨酸、半胱氨酸引起,因此样品中若含有酚类、柠檬酸和巯基化合物,均有干扰作用。
2、福林酚仅在酸性条件下稳定,所以加入试剂乙后要立即混匀,在试剂乙被破坏之前完成反应,否则显色程度减弱。
3、用该方法测定蛋白浓度会受到蛋白质的特异性影响,即不同蛋白质因络氨酸、色氨酸含量的不同而使显色强度稍有不同,标准曲线也不是严格的直线形式。
The Folin–Ciocalteu reagent (FCR) or Folin's phenol reagent or Folin–Denis reagent, also called the gallic acid equivalence method (GAE), is a mixture of phosphomolybdate and phosphotungstate used for the colorimetric in vitro assay of phenolic and polyphenolic antioxidants. It is named after Otto Folin, Vintilă Ciocâlteu, and Willey Glover Denis.
The reagent does not measure only phenols, but will react with any reducing substance. It therefore measures the total reducing capacity of a sample, not just phenolic compounds. This reagent is part of the Lowry protein assay, and will also react with some nitrogen-containing compounds such as hydroxylamine and guanidine. The reagent has also been shown to be reactive towards thiols, many vitamins, the nucleotide base guanine, the trioses glyceraldehyde and dihydroxyacetone, and some inorganic ions. Copper complexation increases the reactivity of phenols towards this reagent.
This reagent is distinct from Folin's reagent, which is used to detect amines and sulfur-containing compounds.
Because it measures anti-oxidant capacity in vitro, the reagent has been used to assay foods and supplements in food science. The oxygen radical absorbance capacity (ORAC) used to be the industry standard for antioxidant strength of whole foods, juices and food additives
