产品介绍:
油性红 O(OiRed O)是一种脂溶性偶氮染料,是很强的脂溶剂和染脂剂,能特异性地使组织和细胞内中性甘油三脂、脂质以及脂蛋白等染色。当组织切片置入染液时,染料则离开染液而溶于组织内的脂质(如脂滴)中,使组织内的脂滴呈红色。用于分析细胞样品中脂质状况。本产品为饱和油红O染色液,操作简捷,性能稳定,着色清晰。本制品仅限于实验试科研使用。

操作说明:(仅供参考)

1. 工作液配置:饱和油红O原液按 3:2(油红 0:蒸馏水)加入蒸馏水,混匀,室温放置 5-10min,过滤后使用。

2. 染色程序
①切片用甲醛-钙固定 10 分钟。
②蒸馏水充分洗涤,60%异丙醇浸洗。
③油红O染液染色 10 分钟(染液可回收再利用)。
④60%异丙醇分化至间质清晰,蒸馏水洗。
⑤Mayer 苏木素复染 3-5 分钟,蒸馏水洗。
⑥甘油明胶封片,镜检。

染色结果:
脂滴 橙色到鲜红色
细胞核 蓝色
间质 无色

注意事项:
1.为了您的安全和健康,请穿实验服并戴一次性手套操作。
2.油红O染色时应避免试剂挥发过多,否则易形成背景沉淀。染色时染色液应充分覆盖组织细胞。
3.60%异丙醇分化,可在镜下控制至脂肪组织呈鲜红色,间质无色时为度。建议使用苏木素复染,增加染色对比度。

4.第一次使用本试剂时建议先取 1-2个样品做预实验。


Oil Red O (Solvent Red 27, Sudan Red 5B, C.I. 26125, C26H24N4O) is a lysochrome (fat-soluble dye) diazo dye used for staining of neutral triglycerides and lipids on frozen sections and some lipoproteins on paraffin sections.   It has the appearance of a red powder with an absorbance maximum at 518 nanometers.


Product Introduction:
Oily Red O(OiRed O) is a lipid soluble azo dye that is a strong lipid solvent and stain that can specifically stain neutral triglycerides, lipids, and lipoproteins in tissues and cells. When the tissue section is placed in the dye solution, the dye leaves the dye solution and dissolves in the lipids (such as lipid droplets) in the tissue, making the lipid droplets in the tissue red. For the analysis of lipid status in cell samples. This product is saturated oil red O dyeing liquid, simple operation, stable performance, clear coloring. This product is for experimental research use only.


Operating Instructions :(for reference only)

1. Working liquid configuration: The saturated oil red O stock liquid is 3:2(oil red 0: distilled water) added to distilled water, mixed well, left at room temperature for 5-10min, filtered and used.

2. Dyeing procedures
① Sections were fixed with formaldehyde-calcium for 10 minutes.
② Fully washed with distilled water and soaked with 60% isopropyl alcohol.
③ Oil red O dye for 10 minutes (the dye can be recycled).
④60% isopropyl alcohol differentiated to clear interstitium, washed with distilled water.
⑤ Redye Mayer hematoxylin for 3-5 minutes and wash with distilled water.
⑥ Glycerin gelatin seal, microscopic examination.

Dyeing result:
Grease drops orange to bright red
Nuclear blue
Achromatic interstitium

Note:
1. For your safety and health, please wear a lab coat and disposable gloves.
2. Oil red O staining should avoid excessive volatilization of the reagent, otherwise it is easy to form background precipitation. The staining solution should fully cover the tissue cells during staining.
3.60% isopropyl alcohol differentiation, which can be controlled under the microscope until the adipose tissue is bright red and the interstitial is colorless. It is recommended to use hematoxylin to increase the staining contrast.
4. When using this reagent for the first time, it is recommended to take 1-2 samples for pre-test.
