解离液常用于根尖等细胞的涂片，它能使细胞壁中层（果胶质）溶解，而使细胞分开，便于最后制片时能被压成一薄层进行显微观察。常用于植物细胞的有丝分裂和DNA的粗提与鉴定。

HCL水解时间对染色效果影响很大，水解时间短，易着色但较难压片；水解时间长，染色慢而色淡；超过20min或水解温度过高，往往染色极淡，或染不上色.各种材料最合适的时间有差别，一般来说，大染色体材料水解时间可较长，小染色体材料宜短。

解离就是用要药液使组织的细胞相互分离开来，便于最后制片时能被压成一薄层进行显微观察。解离液中酒精的作用是迅速杀死细胞，固定细胞的分裂相（将洋葱根尖剪下后，如果不立即投入固定液中将其杀死并令其各种细胞结构凝固，那么，整个根尖将一个分裂期细胞也找不到！）；而HCL能溶解果胶，从而使洋葱细胞的细胞壁软化，并使细胞间的中胶层物质溶解，从而达到分离细胞的目的。另外解离时间不宜过短，否则根尖未充分解离，压片时细胞分不开；但又不能太长，否则使根尖过分酥软，无法进行漂洗和染色，且染色体成分被破坏。这一步是实验成功与否的关键。

但观察动物细胞有丝分裂中，不能用盐酸和酒精来“解离”动物组织。动物细胞不分散的原因是细胞膜上的某些蛋白质分子嵌入到相邻细胞的细胞膜内，有的还形成凹凸式镶嵌结构。盐酸和酒精都不能破坏这两种结构，因此无法用盐酸和酒精来“解离”动物组织。只能用胰蛋白酶，对于植物细胞，胰蛋白酶也没法发挥作用。
