在组织切片过程中，一些组织内含有骨质或钙化灶时，含钙的组织不宜直接用石蜡包埋切片。这是因为钙和石蜡之间的密度不同，较难切出完整的切片。对含钙组织最好固定之后，再进行脱钙或二者同时进行。然后进行下游操作如脱水、透明、浸蜡、包埋、切片。用于脱钙的试剂很多，脱钙剂包括有机酸、无机酸、乙二胺四乙酸（EDTA)以及电解法脱钙。在有机酸中，甲酸、乙酸比较适合于骨髓组织脱钙。

5%快速酸性脱钙液含甲醛可部分地抑制Xiao酸的浸软作用，这既具有脱钙作用，又具有固定作用，Xiao酸脱钙迅速，组织不膨胀，掌握好脱钙时间可直接脱水，可不影响染色效果。但最好先洗去Xiao酸。一般于1-3d内脱钙。

用此溶液作脱钙液的缺点是形成亚硝酸而使溶液呈黄色，产生染色即迅速影响脱钙速度，且组织的黄染可能会妨碍随后的染色反应。在溶液内加入0.1%尿素可以防止变黄而使之无色，但尿素仅有暂时作用，一旦溶液显淡黄时需要加入。此溶液对组织的酸化作用较快，非紧急脱钙一般不用。


实验原理：

5%快速酸性脱钙液对骨组织能迅速固定的同时又能起到脱水作用 , 在 50~ 60℃ 加热状态下 , 使骨组织中分子运动加快 , 从而加速良脱钙液中硝酸根离子与钙离子结合 , 快速析出骨组织中的钙离子，极大缩短了脱钙的时间。


附录：
脱钙终点的测定（物理法）:采用针刺、手掐、钳夹等方法，当骨组织变软或针刺时没有阻力感即可终止脱钙。物理检测法会对组织结构有一定的损害，尽量避免用力过大或反复检测。

The disadvantage of using this solution as decalcification solution is that the solution is yellow due to the formation of nitrous acid, which will quickly affect the rate of decalcification, and the yellow staining of tissues may hinder the subsequent staining reaction. Adding 0.1% urea to the solution can prevent yellowing and make it colorless, but urea has only a temporary effect and needs to be added once the solution becomes yellowish. This solution acidifies tissue quickly and is generally not used for non-emergency decalcification.


Appendix:
Determination of the end point of decalcification (physical method) : acupuncture, hand pinch, clamp and other methods are used to terminate decalcification when the bone tissue becomes soft or there is no resistance to acupuncture. Physical detection will have certain damage to the organization structure, try to avoid excessive force or repeated detection.
