甲苯胺蓝(Toluidine Blue O)是一种常用的人工合成染料, 属于醌亚胺染料类, 这类染料主要含有胺基和醌型苯环两个发色团，从而成色原显色，甲苯胺蓝中的阳离子有染色作用, 组织细胞的酸性物质与其中的阳离子相结合而被染色。甲苯胺蓝还含有两个助色团，能促使染料产生电离成盐类, 帮助发色团对组织产生染色力, 使切片上的组织细胞着色，可染细胞核使之呈蓝色；另外肥大细胞胞质内含有肝素和组织胺等异色性物质，遇到甲苯胺蓝可呈异染性紫红色，实验室科研上常用于肥大细胞染色。

甲苯胺蓝染色液(细胞专用)适用于细胞内酸性黏液多糖染色，尤其适用于含硫的酸性黏液物质染色，染色后酸性黏多糖呈红色。该试剂仅用于科研领域，不适用于临床诊断或其他用途。

操作步骤(仅供参考)：
1、细胞涂片后，立即放入 95%的乙醇中固定 15s，取出放在纸巾上。
2、滴加甲苯胺蓝染色液(细胞专用)进行滴染 5min。
3、滴加等量蒸馏水于涂片上混匀，染色 15min。
4、水洗、晾干、镜检。 染色结果：细胞内酸性黏液多糖呈红色。

注意事项：
1、为了证实染色效果，可选用慢性粒细胞白血病血片(嗜酸性粒细胞)或再生障碍性贫血骨髓涂片(组织嗜酸性粒细胞)作对照。

Toluidine Blue (Toluidine Blue O) is a commonly used synthetic dye, belongs to the quinone imide dye, this kind of dye mainly contains amine group and quinone type benzene ring two hair chromophore, so that the color of the cations in toluidine blue have a dyeing effect, and the acidic substances in the tissue cells are stained by combining with the cations. Toluidine blue also contains two cochromophores, which can promote the dye to ionize into salts, help the chromophores to produce staining force on the tissue, coloring the tissue cells on the section, and staining the nucleus to make it blue; In addition, mast cells contain heterochromatic substances such as heparin and histamine in the cytoplasm, and toluidine blue can be metachromatic purplish red, which is often used for mast cell staining in laboratory research.


Operation steps (for reference only) :
1. Immediately after smear, fix in 95% ethanol for 15s, remove and place on paper towel.
2. Add toluidine blue staining solution (special for cells) for 5min.
3. Add an equal amount of distilled water to the smear, mix it well, and stain it for 15min.
4, wash, dry, mirror inspection. Staining results: Intracellular acidic mucous polysaccharide was red.
